      [摘要]目的 建立问荆总黄酮含量测定方法,并优化其提取工艺。方法 通过单因素试验考察提取温度、超声功率、超声时间和料液比对问荆总黄酮含量的影响;采用Box-Benhnken试验设计原理进行4因素3水平试验设计及响应面分析优化总黄酮提取工艺;运用NaNO2-Al(NO3)3-NaOH比色法测定总黄酮含量。结果 4种因素对问荆总黄酮含量影响排序为料液比>超声功率>超声时间>提取温度。优选得到总黄酮最佳提取工艺为超声时间55 min、料液比1∶40 g/ml、提取温度40℃、超声功率60%(最大功率1000 W)。在此条件下,测得问荆营养茎、繁殖茎时期总黄酮含量分别为13.32、6.92 mg/g。结论 本实验所建立的总黄酮含量测定方法及提取工艺,操作简捷易行,提取效率高,稳定性好,适用于问荆总黄酮的提取和含量测定。

      [Abstract] Objective To establish a method for the determination of total flavonoids in Equisetum arvense L. and optimize its extraction process. Methods The effects of extraction temperature, ultrasound power, ultrasound time, and material-liquid ratio on the total flavonoids in Equisetum arvense L. were investigated by single factor experiments. Box-Benhnken experimental design principle was used to optimize the extraction process of total flavonoids by 4 factors and 3 levels experimental design and response surface analysis. The content of total flavonoids was determined by NaNO2-Al(NO3)3-NaOH colorimetry. Results The order of influence of four factors on the content of total flavonoids in Equisetum arvense L. was material-liquid ratio > ultrasound power > ultrasound time > extraction temperature. The optimum extraction process of total flavonoids was as follows: ultrasound time as 55 min, material-liquid ratio as 1∶40 g/ml, extraction temperature as 40℃ and ultrasound power as 60% (maximum power as 1000 W). Under these conditions, the contents of total flavonoids in the vegetative stem and reproductive stem of Equisetum arvense L. were 13.32 and 6.92 mg/g, respectively. Conclusion The determination method and extraction process of total flavonoids established in this experiment are simple and easy to operate, with high extraction efficiency and good stability, and are suitable for the extraction and content determination of total flavonoids in Equisetum arvense L.
