      [摘要] 目的 采用超高效液相色谱-质谱联用技术(UPLC-MS)检测森林脑炎(TBE)患者外周血清,筛选得到特征性代谢物,用于TBE患者早期筛查和诊断试验。 方法 采用UPLC-MS平台,对2018年4—12月内蒙古林业总医院神经内科住院的30例TBE患者及30例同期体检健康者进行对比分析,结合多元统计分析方法筛选与鉴定特征代谢标志物,统计分析所得特征代谢物对TBE的诊断效能。 结果 共筛选出47种代谢离子,最终鉴定出12种特征代谢物,共有5种特征代谢物表达下降,其中3种代谢物区分效能较高,另有7种表达上升、其中6种区分效能较高。 结论 利用UPLC-MS技术平台检测TBE患者血清特征代谢物是一种有效的方法,其确定的植物鞘胺醇、前列腺素E2乙醇酰胺、胆固醇硫酸盐3种特征代谢物或可成为TBE前期筛查、诊断的检测标准。

      [Abstract] Objective To screen for characteristic metabolites for early screening and diagnostic tests in patients with tick-borne encephalitis (TBE) using ultra-performance liquid chromatography-mass spectrometry (UPLC-MS). Methods The UPLC-MS platform was employed to compare and analyze 30 patients with TBE hospitalized in the Department of Neurology of Inner Mongolia Forestry General Hospital from April to December 2018 and 30 healthy individuals with physical examination during the same period, combined with multivariate statistical analysis methods to screen and identify characteristic metabolic markers, and statistically analyze the diagnostic efficacy of the resulting characteristic metabolites for TBE. Results A total of 47 metabolic ions were screened, and 12 characteristic metabolites were eventually identified. A total of five characteristic metabolites exhibited decreased expression, three of which were highly effective at differentiation. In addition, there was an increase in the expression of seven characteristic metabolites, six of which were distinguished with high efficacy. Conclusion The use of the UPLC-MS platform for the detection of characteristic metabolites in the serum of TBE patients is an effective method, and the identification of three characteristic metabolites, phytosphingosine, prostaglandin E2 ethanolamide and cholesterol sulfate, may become a standard for pre-screening and diagnosis of TBE.
