[摘要] 目的 探讨夏天无治疗胃癌的主要作用机制。 方法 通过中药系统药理学数据库(TCMSP)和GeneCards数据库筛选夏天无治疗胃癌的潜在作用靶点及对应的有效成分;利用Cytoscape 3.7.2软件构建药理网络;将作用靶点导入通过STRING数据库和Cytoscape 3.7.2软件进行蛋白-蛋白互作可视化分析,进一步筛选关键基因;关键基因分别通过DAVID在线平台和GEPIA 2数据库进行富集(GO和KEGG)分析和胃癌预后分析。 结果 夏天无治疗胃癌的有效成分有6种,对应65个靶点与胃癌相关,其中关键靶点45个,它们涉及的信号通路包括钙信号通路、神经活性配体-受体相互作用等;45个关键靶点中有8个靶基因(ADRA1A、ADRA1B、AR、ESR1、F7、OPRD1、PTGS2、RXRA)与胃癌预后相关(P < 0.05)。 结论 本研究通过网络药理学探索出夏天无治疗胃癌的潜在有效成分和靶点以及其作用机制,为夏天无中药的抗癌新作用提供理论依据。 |

[Abstract] Objective To investigate the main function mechanism of rhizoma corydalis decumbentis in treating gastric cancer. Methods Through Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP) and GeneCards database, the potential function targets and corresponding effective components of rhizoma corydalis decumbentis in treating gastric cancer were screened. The pharmacological network was constructed by using the Cytoscape 3.7.2 software. The function target was introduced into the STRING database and Cytoscape 3.7.2 software for visual analysis of protein-protein interaction, and the key genes were further screened. The key genes were enriched (GO and KEGG) and the prognosis of gastric cancer was analyzed by DAVID online platform and GEPIA 2 database respectively. Results There were 6 kinds of effective components of rhizoma corydalis decumbentis in treating gastric cancer, and 65 targets were related to gastric cancer, of which 45 were key targets. The signaling pathways involved included calcium signaling pathway, neuroactive ligand-receptor interaction, etc. A total of 8 of the 45 key targets (ADRA1A, ADRA1B, AR, ESR1, F7, OPRD1, PTGS2, RXRA) were related to the prognosis of gastric cancer (P < 0.05). Conclusion The potential effective components, targets and function mechanism of rhizoma corydalis decumbentis in treating gastric cancer are explored through network pharmacology in this investigation, thereby providing theoretical basis for the new anticancer effect of rhizoma corydalis decumbentis as Chinese materia medica. |
