      [摘要] 目的 基于网络药理学探讨熟地黄-山茱萸治疗2型糖尿病(T2DM)的机制。 方法 首先对熟地黄及山茱萸的有效活性成分进行筛选,其次运用DisGeNET、OMIM以及GeneCards数据库,对熟地黄以及山茱萸的有效成分靶点和T2DM的作用靶点进行预测。将所获得的数据输入在线维恩平台获得疾病与成分的交集靶点,然后将交集靶点导入Cytoscape软件进行可视化分析,获得疾病-成分-靶点网络图,最后进行GO通路的富集分析和KEGG功能注释分析。 结果 筛选出熟地黄以及山茱萸有效活性成分共54种。收集T2DM的作用靶点共450个,相互交集的靶点共24个。GO富集分析发现主要富集在细胞成分等方面,生物学过程主要富集在对基因的调节、调控。KEGG通道路径富集分析发现其涉及多条通路,如营养信号通路、胰岛素抵抗、神经活性配体-受体相互作用等。 结论 初步发现了熟地黄-山茱萸治疗T2DM的分子机制。

      [Abstract] Objective To investigate the mechanism of radix rehmanniae praeparata and cornus officinalis in the treatment of type 2 diabetes mellitus (T2DM) based on network pharmacology. Methods Firstly, the effective active components of radix rehmanniae praeparata and cornus officinalis were screened. Secondly, the effective components target of radix rehmanniae praeparata and cornus officinalis and the action target of T2DM were predicted by using DisGeNET, OMIM and GeneCards databases. The obtained data were put into the online Venn platform to obtain the intersection targets of diseases and components. Then the intersection targets were imported into Cytoscape software for visual analysis to obtain the network diagram of diseases-components-targets. Finally, the enrichment analysis of GO pathway and KEGG function annotation analysis were carried out. Results A total of 54 kinds of effective active components from radix rehmanniae praeparata and cornus officinalis were screened. A total of 450 action targets of T2DM were collected, and there were 24 intersecting targets. GO enrichment analysis showed that it was mainly concentrated in cell components, and the biological process was mainly concentrated in the regulation and control of genes. Enrichment analysis of KEGG channel pathway showed that it involved multiple pathways, such as nutritional signaling pathway, insulin resistance, and neuroactive ligand-receptor interaction. Conclusion The molecular mechanism of radix rehmanniae praeparata and cornus officinalis in the treatment of T2DM was preliminarily discovered.
