      [摘要] 目的 探讨大黄素缓解类风湿关节炎(RA)痛的作用及机制。 方法 将30只C57BL/6小鼠随机分为对照组、模型组和大黄素给药组;采用左后膝关节腔注射完全弗式佐剂构建小鼠RA痛模型;腹腔注射大黄素进行给药;记录小鼠痛觉行为及运动能力改变;免疫印迹法(Western blot)检测各组动物脊髓组织氧化及炎症相关蛋白变化。 结果 完全弗式佐剂注射14 d后,与对照组小鼠相比,模型组小鼠自发缩足次数增加,机械痛阈值下降,运动能力减弱,脊髓组织核因子κB(NF-κB)、白介素1β(IL-1β)、NOD样受体热蛋白结构域相关蛋白3(NLRP3)、小胶质细胞标记蛋白游离钙结合适配器蛋白1(Iba1)、环氧化酶2(COX-2)蛋白水平均增加;核因子E2相关因子2(Nrf2)蛋白表达降低,差异均有统计学意义(P < 0.05)。大黄素腹腔给药后,与模型组小鼠相比,小鼠自发缩足次数减少,机械痛阈值增加,改善小鼠的运动能力,降低NF-κB、IL-1β、NLRP3、Iba1和COX-2蛋白表达水平,升高Nrf2蛋白表达水平,差异均有统计学意义(P < 0.05)。 结论 大黄素具有显著缓解RA痛的作用效果,其机制是通过抑制脊髓炎症和氧化应激反应。

      [Abstract] Objective To investigate the efficacy and mechanism of emodin in relieving rheumatoid arthritis pain. Methods A total of 30 C57BL/6 mice were selected and randomly divided into the control group, the model group and the emodin administration group. The rheumatoid arthritis pain model of mice was established by injecting Complete Freund’s Adjuvant (CFA) into the left posterior knee joint cavity. Emodin was injected intraperitoneally for administration. The changes of pain perception behavior and motor ability of mice were recorded. Western blot was used to detect the changes of oxidation and inflammation-related proteins in the spinal cord tissues of animals in each group. Results After 14 days of CFA injection, compared with the mice in the control group, the number of spontaneous foot contraction of the mice in the model group increased, the threshold of mechanical pain decreased, and the motor ability weakened. Meanwhile, the nuclear factor-κB(NF-κB), the interleukin-1β(IL-1β), the NOD-like receptor thermal protein domain associated protein 3 (NLRP3), the microglia marker protein ionized calcium binding adapter protein 1(Iba1) and the cyclooxygenase 2 (COX-2) in spinal cord tissues all increased. The expression of nuclear factor E2-related factor 2 (Nrf2) protein decreased, with statistically significant differences (P < 0.05). After intraperitoneal administration of emodin, compared with the mice in the model group, the number of spontaneous foot contractions of the mice decreased, the threshold of mechanical pain increased, the motor ability of mice improved, the expression levels of NF-κB, IL-1β, NLRP3, Iba1 and COX-2 proteins decreased, and the expression level of Nrf2 protein increased, with statistically significant differences (P < 0.05). Conclusion Emodin can significantly relieve rheumatoid arthritis pain, with its mechanism of inhibiting spinal cord inflammation and oxidative stress reaction.
