      目的 探讨姜黄素对 β- 淀粉样蛋白损伤神经干细胞(NSC)增殖和凋亡的影响。 方法 选取 NSC 分为对照组、姜黄素组、Aβ 组和观察组,其中姜黄素组、Aβ 组和观察组分别加入终浓度为 5μmol/L 的姜黄素、0.5μmol/L 的 Aβ+DMSO(10μL)、5μmol/L 的姜黄素 +0.5μmol/L 的 Aβ,对照组加入 DMSO(10μL)。使用姜黄素治疗 NSC,通过细胞计数法计算 NSC 的增殖速度,噻唑蓝 (MTT) 测定 NSC 活性,逆转录。聚合酶链反应 (RTPCR) 测定 NSC 中 Nestin mRNA 及凋亡相关基因 Caspase3 mRNA 表达,免疫组化测定 NSC 向神经元和星形胶质细胞的分化潜能。 结果 与对照组比较,姜黄素组 NSC 的细胞活力、Nestin mRNA 表达明显增加,Caspase3 mRNA 表达明显降低(P < 0.05 或 P < 0.01),而 Aβ 组和观察组 NSC 活力、Nestin mRNA 表达和 Tuj-1、GFAP明显降低,Caspase3 mRNA 表达明显增加,差异有统计学意义(P < 0.05 或 P < 0.01)。与 Aβ 组比较,观察组NSCs 细胞活力、Nestin mRNA 表达和 Tuj-1、GFAP 明显增加,Caspase3 mRNA 表达明显降低,差异有统计学意义(P < 0.05 或 P < 0.01)。 结论 姜黄素促进 NSC 增殖,促进 NSC 向神经元分化,逆转 Aβ 对 NSC 的损伤可能是由于抑制了凋亡基因表达。

      Objective To investigate the effects of curcumin on the proliferation and apoptosis of neural stem cells (NSC) injured by beta-amyloid protein. Methods NSCs were divided into control group, curcumin group,Aβgroup and observation group. Curcumin group, Aβgroup and observation group were added curcumin with final concentration of 5μmol/L, 0.5μmol/L of Aβ+ DMSO (10μL), 5μmol/L of curcumin + 0.5μmol/L of Aβrespectively, and DMSO (10μL) was added in control group. Curcumin was used to treat NSC. The proliferation rate of NSC was calculated by cell counting method. The activity of NSC was measured by MTT and the reverse transcription was performed. The expression of Nestin mRNA and apoptosis-related gene Caspase3 mRNA in NSC was detected by polymerase chain reaction (RT-PCR), and the differentiation potential of NSC to neurons and astrocytes was determined by immunohistochemistry. Results Compared with the control group, the cell viability and Nestin mRNA expression of NSC in curcumin group increased significantly, Caspase3 mRNA expression decreased significantly (P < 0.05 or P < 0.01), while NSC activity, Nestin mRNA expression and Tuj-1, GFAP in Aβ group and observation group decreased significantly, Caspase3 mRNA expression increased significantly, the difference was statistically significant(P < 0.05 or P < 0.01). Compared with Aβ group, NSCs cell viability, Nestin mRNA expression, Tuj-1, GFAP in the observation group were significantly increased, while Caspase3 mRNA expression was significantly decreased, the difference was statistically significant (P < 0.05 or P < 0.01). Conclusion Curcumin promotes the proliferation of NSC and the differentiation of NSC into neurons. Reversing the damage of Aβ to NSC may be due to inhibiting the expression of apoptotic genes
